I know you might be wondering what this is all about right?? I know most of us on one or two occasions have probably said they are used to the intake of paracetamol and that it has no effect on them anytime we take it in to gain relief of pain.

Most times, we tend to seek other drugs probably more potent or has better efficacy than that which we are already taking. This is one common issue mostly faced by patients but then I think it's best you know what is going on in your body before you end up complicating things.

When drugs are taken into the body they mostly bind to receptors and these receptors undergo conformational changes to transduce the signal to the effector organ to elicit an effect that we see on the outside.

These receptors are are selective and specific to ligands(drugs) and there are times when drugs may fail to bind to these receptors as a result of the affinity of the drug to the receptor or body conditions which could impede on drug recptor intereations.

When this happens people tend to take an extra dose of the medication which then leads to toxicity in the body since the already taken drug is still flying around and hasn't been bonded to any recptor or been cleared out by the liver which then leads to increased plasma levels of the drug leading to toxicity.

Our body has also been designed in a way that when there is increased concentration of a drug, our receptors have a defense mechanism where it can be insensitive to these drugs coming its way and therefore no effect would be seen after even taking extra doses of the drug.

Sometimes too the receptor undergoes endocytotis and is sequestered by the body cells and makes no receptors available for drug binding to elicit an effect .

So you might be wondering what is going on your body when you take a drug when no effect is seen . There are other reasons to why this might be experienced such as 1st pass metabolism and several other which shall be spoken on in my next posts. But then in a nut shell this is a fair idea about what happens when it comes to drug-receptor binding.
