Whoa! 2nd week of this course. Last week I knew Hematopoiesis . Thanks dexsyluz dear for conducting a course on this subject. Now it's all about Hemoglobin. I think every person needs to know about this.

Death of Rafael Rangel


Rafael Rangel died from consuming of Drabkin's reagent. He has ingested Drabkin's reagent or injected it into the body in some way. A few moments after entering the reagent, the following processes were completed, resulting in the death of Rafael Rangel.
Drabkin's reagent is a solution primarily used by laboratories to measure hemoglobin levels in blood samples collected in the laboratory. It has two types of components.

  • Potassium ferricyanide and
  • Contains potassium cyanide.

When anticoagulated blood comes into contact with Drabkin's reagent, the blood cells break down and release hemoglobin. Then Potassium ferricyanide converts blood hemoglobin to methemoglobin, which can be measured by color. Potassium cyanide, the main ingredient of the Drabkin's reagent. Itis highly toxic. We know that all cyanides are highly toxic enough to kill people in a short time. If potassium cyanide is ingested in sufficient amounts, it can inhibit cellular respiration by inhibiting cytochrome c oxidase in the mitochondria of cells, resulting in a failure of cells to use oxygen. And without enough oxygen, everything changes quickly. Initially due to lack of oxygen the person becomes suffocated, senseless and ultimately dies.


Describe Hemoglobin


The word hemoglobin is derived from the combination of two Greek words. Heme means blood and Globin means protein. Among the three blood cells of animals, the protein present in red blood cells is called globin, so we can say that hemoglobin is found in red blood cells of animals. It is an iron conjugated protein. All vertebrates and invertebrates, including humans, have hemoglobin in their blood (except fish species) whose main function is to transport oxygen from the lungs to tissues and organs. It also plays an important role in returning carbon dioxide from the tissues to the lungs.

There are two basic components of hemoglobin.

  • Hemo Group
  • Globin chains

Normal adult hemoglobin (hemoglobin A) is composed of two alpha (α) chains and two beta (β) chains. Each heme group contains iron that can bind a molecule of oxygen, allowing hemoglobin to carry four oxygen molecules. The protein helps maintain blood pH and contributes to blood flow and blood pressure regulation.

Hemoglobin helps in maintaining blood density.
Blood is red because of the presence of hemoglobin. Only red blood cells are red due to the presence of hemoglobin, the rest of the blood cells are colorless due to the absence of hemoglobin.


Automated Hematocrit vs Manual Hematocrit

This method provides quick results. It provides values ​​of a complete blood count within 2 minutesonly Delivering, processing and analyzing results takes longer.
This method is used most of the time in the laboratory as it is easier to use.This method isnt used most of the time in the laboratory.
It does not help in basic skill development in automatic air so this method is not taught in universitiesThis method is taught in universities because it helps in basic skill development without being automatic.
Reduces human error and variabilityin manual can lead to human error.
Can process multiple samples simultaneouslyCannot process multiple samples at once.
Being automatic, it requires relatively expensive equipment and maintenance.It is less expensive than the automated method.
Anticoagulant and Drabkin's reagent arent required for automated determination always (often use in few specific methods).Anticoagulant and Drabkin's reagent are required for manual determination.

Explain a real-life example with the concept of hematocrit

When it is about the concept of hematocrit then it is the parsentage of red boold cell volume in total blood cell. It could be high or low and normal. In our real life we often asked to test our hematocript by our doctor. A relative of mine went to the doctor for some problem and the doctor recommended some blood tests. Where his hematocrit test is done. This is done as part of a complete blood count (CBC). As I said before, through CBC, the amount of red blood cells in the blood can be understood whether it is low, high or normal. What the doctor tells us from his complete test is that he has a low hematocrit value and a low hematocrit value means that the proportion of red blood cells in the blood is lower than normal. He has very few healthy red blood cells in his blood. This condition is called anemia. Then the doctor suggested him a medicine called "Folison" and prohibited him from consuming foods containing extra iron. From practical experience I learned about low hematocrit levels and later as part of this course I learned that high hematocrit levels are due to dehydration.

Hi! Cyanide is a highly toxic substance, I read that it is often widely used to have the same end as the doctor, something that completely surprised me since although it is quite fast acting, it's very painful!
